Abstract
Objective: To study the collagen structure and antioxidant activity of traditional Chinese medicine elephant skin and its intended substitutes, pig skin and fish scales. Methods: Through the orthogonal experimental design method, the optimal extraction conditions of elephant skin collagen were optimized. The structure and content of elephant skin collagen were compared with those of the proposed substitute collagen. The salicylic acid method was used to determine their respective hydroxyl radical scavenging abilities. Results: For the first time, collagen with a complete structure and high quality was extracted from elephant skin. The structural type of collagen is similar to that of the proposed substitute. The IC50 of the free radical scavenging ability of elephant skin collagen, pig skin collagen, and fish scale collagen were 0.51 respectively. 0.60,0.42g¡¤L-1. Conclusion: Through comparison and identification, elephant skin collagen is type I collagen with strong antioxidant capacity. The active ingredients of elephant skin are clarified, which provides an important reference for further research on elephant skin substitutes in the future.
