Abstract
Natural collagen was extracted from grass carp skin, and was confirmed to be type I collagen with a complete triple helix molecular structure by DSC, circular dichroism and SDS-PAGE analysis. After heat treatment of the collagen under different conditions, low-denaturation, high-denaturation and completely-denatured collagen samples were obtained respectively. The thermal denaturation enthalpy measurement results showed that the thermal denaturation degrees were: 16.9%, 57.9% and 100% respectively. On this basis, we focused on studying the changes in collagen structure and collagen sponge material properties after thermal denaturation treatment. Experimental results show that the starting temperature of grass carp skin collagen denaturation (To, 33¡ãC) is the threshold temperature for changes in its molecular structure and collagen sponge material properties. A brief heating history above this temperature can lead to partial or complete dissociation of the collagen triple helix structure. At the same time, as the degree of collagen denaturation increases, the orderliness of its material structure, material mechanical properties and resistance to enzymatic hydrolysis also decrease, but its ability to promote cell proliferation increases. The above research results confirm that lower than the starting temperature of thermal denaturation of collagen is a safe temperature condition for natural collagen extraction and collagen material application.
