Abstract
Using tilapia tail as raw material, we studied the removal method of fish tail fat-soluble pigment and the extraction process of collagen, measured the purity of collagen, and identified the type of collagen. The results show that the best solution for pigment removal is a mixed solvent of chloroform-methanol-water (1:2:0.8, V/V), a solid-to-liquid ratio of 1:15 (g/mL), and a time of 36 hours. Under these conditions, the pigment removal rate Reaching 2.85% of the wet mass of the fish tail; orthogonal experiments determined that the optimal process for collagen extraction is guanidine hydrochloride concentration 3mol/L, pepsin addition amount 3.0% (based on crude protein amount), time 72h, and solid-liquid ratio 1: 50 (g/mL) and extraction times 2 times. Under these conditions, the collagen extraction rate reached 4.96% of the wet mass of the fish tail, accounting for 40.39% of the crude protein content of the fish tail; the collagen content in the extract reached 94.63%, and it showed Characteristics of Type I Collagen
