Abstract
This article uses chicken articular cartilage as raw material to prepare type II collagen. Different concentrations of pepsin were added under acidic conditions to enzymatically hydrolyze the telopeptides of the three-stranded helix chain of type II collagen in chicken articular cartilage, converting type II collagen from insoluble to soluble. The changes in substrate viscosity with enzymatic hydrolysis time were studied to determine the best Enzyme concentration and enzymatic hydrolysis time. Type II collagen was purified from enzymatic hydrolysis samples using deae-sephalosecl6b ion exchange column and repeated washing with deionized water. Research shows that the optimal addition concentration of pepsin is 1% (w/w) and the optimal enzymatic hydrolysis time 24h. The extraction rates of type II collagen by the two purification methods were 67% and 81% respectively. The results of sds-page electrophoresis, high-performance liquid chromatography, amino acid composition analysis and ultraviolet absorption spectrometry confirmed that type II collagen prepared from chicken articular cartilage has high purity.
