Abstract
Using grass carp skin and snakehead skin as raw materials, acid-soluble collagen and enzyme-soluble collagen were extracted, separated and purified. On this basis, the two types of collagen were compared through structural identification, thermal stability analysis and in vitro enzyme degradation performance analysis system. The similarities and differences in molecular structure and physical and chemical properties of collagen are compared with those of porcine skin collagen from mammalian sources. The results show that the five extracted collagens are all typical type I collagen; collagens from different extracted sources have similar secondary structures, but there are still significant differences in amino acid composition, protein primary structure, etc. Differential scanning calorimeter (DSC) analysis results show that the thermal denaturation temperatures of the five types of collagen are: pig skin enzyme-soluble collagen (PSC) 41.58¡ãC, grass carp acid-soluble collagen (ASC) 35.62¡ãC, grass carp skin PSC 35.81¡æ, snakehead skin ASC 33.85¡æ and snakehead skin PSC 34.06¡æ; the analysis results of in vitro enzyme degradation performance show that the enzymatic degradation rate of several collagen samples is in the order of snakehead skin PSC> grass carp skin PSC> blackhead fish Skin ASC> Grass carp skin ASC> Pig skin PSC. Compared with grass carp skin and snakehead skin collagen, pig skin collagen has better thermal stability and resistance to enzyme degradation. Between the two collagens derived from freshwater fish, grass carp skin collagen has better thermal stability and enzymatic stability than snakehead skin collagen. At the same time, collagen extracted by acid method has better enzymatic stability than collagen extracted by enzymatic method.
