Abstract
Total internal reflection infrared spectroscopy was used to study the amide I band of collagen and collagen-rich suckling rat skin before and after glutaraldehyde treatment. The results show that after treatment with glutaraldehyde, the area percentage of the higher wave number component among the three main spectral components located around 1660, 1643, and 1633 cm-1 in the amide I band of the infrared spectrum of collagen and neonatal mouse skin decreased, while the area percentage of the higher wave number component located at The component area percentage increases on the intermediate and low wavenumber sides. This change is similar to the changing trend of these three spectral components after thermal denaturation of collagen [1], suggesting that glutaraldehyde treatment weakens the hydrogen bonds between collagen triple helices and increases the spacing between helices. The enlargement of the spaces between helices provides possible sites for collagen calcification.
