Abstract
Objective: To study the degradation process of collagen and key unit operations in the donkey hide gelatin production process. Methods: Samples were taken during the production process of donkey-hide gelatin and its amino acid composition and relative molecular mass range were analyzed. High-performance liquid chromatography mass spectrometry was used to analyze its peptide composition. Results: During the production process of donkey-hide gelatin, the glycine content in the raw gelatin increased by 16.8%, indicating that the collagen content in the raw gelatin gradually increased; the gel filtration chromatography analysis results showed that the relative molecular weight of the raw gelatin gradually decreased, and the degradation process of collagen was mainly concentrated in Normal pressure concentration stage; mass spectrometry analysis results show that in addition to collagen, there are degradation products of extracellular matrix proteins and some cytoskeletal proteins in the raw gel. The substances removed in the form of foam during the normal pressure concentration process are mainly non-collagen proteins. of degradation products. Conclusion: The degradation process of collagen in the donkey-hide gelatin production process is mainly concentrated in the atmospheric pressure concentration stage. The atmospheric pressure concentration process is a key unit operation that affects the composition of donkey-hide gelatin products.
