Experimental Study On The Preventive And Therapeutic Effects Of Sika Deer Antler Collagen Enzymatic Hydrolyzate On Retinoic Acid-Induced Osteoporosis In Rats

Abstract
Objective: To observe the effect of sika deer velvet collagen enzymatic hydrolyzate (CSDV) on retinoic acid-induced osteoporosis in rats. Methods: 50 white rats were randomly divided into 5 groups: control group, model group and CSDV high, medium and low dose groups (0.4, 0.2, 0.1g¡¤kg-1¡¤d-1). Rat osteoporosis model was established by intragastric administration of 70 mg¡¤kg-1¡¤d-1 of retinoic acid for 14 days. The animals were sacrificed after 30 days of treatment with velvet antler collagen enzymatic hydrolyzate by gavage while making the model. Collect blood to separate serum, and measure serum calcium, phosphorus, and alkaline phosphatase contents; take the left femur and weigh it, calculate the bone weight coefficient, and measure bone biomechanical strength; take the right femur and do bone tissue morphometric measurement. Results: Compared with the model group, CSDV could significantly improve the decrease in bone density, increase in serum alkaline phosphatase, and decrease in calcium and phosphorus content in rats induced by retinoic acid; improve obvious changes in bone pathological morphology; and improve the number of bone trabeculae. Decrease, average trabecular bone width narrows, average trabecular bone spacing becomes larger, bone volume decreases, average bone cortical thickness becomes thinner; bone biomechanical strength indicators improve; femur maximum load decreases, maximum deflection becomes shorter, and bone stress decreases , reduced bone strain and other osteoporotic changes. However, the above functions vary with the dosage of velvet antler collagen enzymatic hydrolyzate. Conclusion: The medium-dose group of velvet antler collagen enzymatic hydrolyzate can increase bone mass, normalize the microstructure of bone tissue, increase bone biomechanical strength, and has a preventive and therapeutic effect on osteoporosis in rats.
