Abstract
This study aims to optimize the extraction method of Peruvian squid skin collagen and characterize its main properties to provide a basis for the high-value utilization of squid skin and the preparation of collagen. The acetic acid combined with pepsin method was used to extract collagen from Peruvian squid skin, and the extraction conditions were optimized through single factor and response surface analysis. The results showed that the addition amount of pepsin was 2% (w/w), the extraction time was 53 hours, and the acetic acid concentration was 0.70 mol¡¤ L-1, solid-liquid ratio 1:240 (w/v), the collagen extraction rate can reach 51.47%, and the collagen is purified through salting out and ultrafiltration. SDS-PAGE results show that squid skin collagen is mainly composed of ¦Á1, ¦Á2 and ¦Â chains, and is type I collagen; amino acid composition analysis shows that it has the highest glycine content (281 residues/1000 residues) and 167 imine residues. base/1000 residues.
