Abstract
Using horse-faced fish skin as raw material, the pepsin extraction process of horse-faced fish skin collagen was optimized, and the structure of the extracted collagen was analyzed to provide an experimental basis for the preparation of horse-faced fish skin collagen. The optimal conditions for extraction were obtained: pepsin plus enzyme amount 16?800?U/g, buffer pH?3.0, solid-liquid ratio 1:30 (g/mL), extraction time 21.5?h, and extraction of horse-faced fish skin collagen. The rate is 27.6%. Amino acid analysis, ultraviolet spectrum analysis, Fourier transform infrared spectrum analysis, and vertical electrophoresis analysis showed that the proposed collagen conforms to the characteristics of type I collagen and maintains the triple helix structure of collagen.
