Abstract
Objective To observe the collagen content of wound granulation tissue in rats with depression combined with skin trauma, and to preliminarily explore the possible mechanism by which depression affects wound healing. Methods According to the modeling method, 126 SD rats were randomly divided into a simple skin trauma group, a skin trauma combined with depression group, and a depression combined with skin trauma group. A hydroxyproline kit was used to detect the hydroxyproline content and picric acid of the wound tissue. After Sirius red staining, the collagen deposition on the wounds of each group at each time point was observed under a polarized light microscope. Results The hydroxyproline content and collagen deposition in the wound granulation tissue of the rats in the depression-combined skin trauma group were continuously lower than those in the other two groups (P<0.01, P<0.05). On the 18th day after injury, the levels in the depression-complex skin trauma group tended to be consistent. Conclusion Depression combined with skin trauma can significantly reduce the synthesis of hydroxyproline in wound tissue and reduce the deposition of collagen fibers, thereby delaying wound healing.
