Abstract
Using Yunnan snapper bones as raw materials, the acid-soluble collagen (ASC) in the fish bones was extracted and prepared by low-temperature acid method, and the amino acid composition, subunit composition, infrared absorption, ultraviolet absorption, and thermal stability of ASC were analyzed. The properties, X-ray diffraction, microstructure, peptide fragments and solubility were comprehensively analyzed. The amino acid composition shows that ASC mainly contains glycine, proline and alanine, while the content of tyrosine, methionine and cysteine is lower; the results of sodium dodecyl sulfate-polyacrylamide gel electrophoresis show that ASC is type I Collagen; ASC has a maximum UV absorption peak at a wavelength of 230 nm; Fourier transform infrared spectrum and X-ray diffraction patterns show that ASC molecules are compactly arranged and maintain its original triple helix structure; differential scanning calorimetry analysis results show The denaturation temperatures of ASC are 86.5 ¡æ and 226.2 ¡æ respectively, and have good thermal stability; scanning electron microscopy shows that ASC molecules are evenly distributed, with a smooth surface and a three-dimensional structure; peptide fragment analysis results show that the amino acid composition of fish bone collagen is mainly Gly -XY, consistent with the characteristics of the primary structure of collagen; when the pH value is less than 4, the solubility of ASC is high. When the mass fraction of NaCl is greater than 4%, the solubility of ASC decreases sharply.
