Type ¢ò Collagen Antagonizes Articular Cartilage Damage Induced By T-2 Toxin

Abstract

Objective To observe the effect of type ¢ò collagen on the early damage of articular cartilage induced by T-2 toxin in rats, and to provide a theoretical basis for exploring the prevention and treatment measures of articular cartilage damage. Methods: 40 Wistar rats were randomly divided into 4 groups, with 10 rats in each group: control group (conventional finished pellet feed), T-2 toxin group (feed containing 100ng/kg T-2 toxin), and type II collagen 0.5 and 5.0 groups. (0.5, 5.0g/L) for 5 consecutive months, the histopathological changes of rat hyaline cartilage were observed under light microscope, and the enzyme-linked immunosorbent assay was used to detect the carboxyl-terminal telopeptide of rat serum type II collagen (CTX-¢ò), Cartilage oligomeric matrix protein (COMP) and urinary pyridinoline (DPD) content. Results Under light microscope, the articular cartilage cells of the rats in the T-2 toxin group were arranged in disorder, the cartilage cells were deformed and degenerated, and large areas of chondrocyte necrosis were seen. However, the low and high dose type II collagen groups showed the formation of fibrils on the cartilage surface and the formation of superficial chondrocytes. Early pathological changes of osteoarthritis include swelling and rounding, reduction of flat chondrocytes, and clustering of chondrocytes. The serum CTX-II and COMP contents of rats in the control group, T-2 toxin group, high-dose and low-dose type II collagen groups were (25.07¡À9.17), (39.17¡À10.49), (28.20¡À9.74), and (29.73¡À9.32) respectively. )¦Ìg/L and (6.38¡À2.23), (24.53¡À4.26), (20.32¡À4.74), (19.44¡À4.92)¦Ìg/L, the DPD contents in rat urine were (4.14¡À1.06), (4.33¡À3.43) respectively ), (5.72¡À3.89), (4.23¡À2.90)¦Ìg/L. Compared with the control group, the serum CTX-II and COMP contents of rats in the model group were significantly increased (all P<0.05), and the DPD content in rat urine had no significant change; compared with the model group, the serum CTX-II and COMP contents of rats in the type II collagen group were significantly increased (all P<0.05). ¢ò. COMP was significantly reduced (all P<0.05), and the DPD content in rat urine had no significant change. Conclusion Type II collagen can antagonize cartilage damage caused by T-2 toxin and delay the destruction process of articular cartilage.
