Abstract
Objective To explore the effect of zaocystype ¢¢ collagen (zc ¢¢) on mesenteric lymph node lymphocyte (mlnls) Treg/th17 cells and cytokines in mice with collagen-induced arthritis (collagen-induced arthritis, cia). Methods Chicken type ¢¢ collagen was injected subcutaneously into the tail root of mice to induce cia model; zc¢¢ was extracted by pepsin digestion method, and high, medium and low doses of zc¢¢ were administered orally. Visual scoring method was used to evaluate joint swelling, and he staining was used to evaluate knee joint histopathology; mouse mlnls was isolated, and the ratio of treg and th17 cells was detected by flow cytometry (fcm); mlnls culture supernatant was detected by enzyme-linked immunosorbent assay (elisa) Activities of TGF-¦Â and IL-17. Results Compared with the normal group, the arthritis and histopathological scores of cia mice were increased (p<0.05), and the ratio of Tregs and Th17 in mlnls and the levels of secreted Tgf-¦Â and IL-17 were increased (p<0.05). Compared with the cia model group, the arthritis and histopathological scores of different doses of zc¢¢ intervention group were reduced (p<0.05), the ratio of Treg cells in mlnls increased, the ratio of th17 cells decreased (p<0.05), and the level of tgf-¦Â secreted by mlnls increased, and secreted il-17 levels decreased (p<0.05). Conclusion Oral administration of zc¢¢ can improve arthritis and histopathological scores in cia mice; zc¢¢ may re-introduce the immune balance of cia mice and alleviate joint inflammation by regulating the treg/th17 ratio and cytokine levels in mlnls.
