Abstract
Objective To observe the adhesion characteristics of epidermal stem cells to type IV collagen at different times. Methods: Human keratinocytes were collected 10, 20, 30, and 60 minutes after attachment, and the cells were counted using a cell counter to calculate their diameters. Adherent cells were identified using ¦Â1 integrin and K19. Flow cytometry was used to detect the positive expression of ¦Â1 integrin, and phase contrast microscopy was used to observe cell morphology. Results As time went by, the number of adherent epidermal stem cells gradually increased. The number of adherent cells in 20 minutes was significantly different from that in 10 minutes (P<0.05), but there was no statistical significance in the number of adherent cells in 30 and 60 minutes. The adherent cells are small and round, with strong reflective ability. The indirect immunofluorescence and immunohistochemistry results of integrin ¦Â1 show that the adherent cells are positive. The positive rate of K19 immunohistochemistry is 31.03%. The positive rate of integrin ¦Â1 measured by flow cytometry. Expression level is 100%. Conclusion IV collagen can quickly adhere to most epidermal stem cells within 20 minutes without affecting cell viability and can be used to sort epidermal stem cells.
