Thermal Denaturation Kinetics Of Enzymatically Soluble Collagen From Jellyfish Fimbriae

Abstract

In order to elucidate the thermal denaturation reaction mechanism of pepsin-solubilized collagen (PSC) in the jellyfish fimbriae, PSC that maintains an intact triple helix structure was used as the research object, and the denaturation temperature of PSC under different heating rates was measured by a microcalorimeter. , as well as the PSC survival rate after heating at 34, 35, 36, 37, 38, and 39¡ãC for different times, and thermal denaturation kinetics analysis was performed. The results show that jellyfish parasol PSC is sensitive to thermal changes. As the heating temperature increases, the amount of heat increased per unit time increases, which accelerates the denaturation rate of jellyfish parasol PSC and shortens the time to complete denaturation. As the heating rate slows down, the absorption rate increases. The thermal peak gradually moves to the low temperature area, that is, the denaturation temperature decreases as the heating rate slows down, but the change in the heating rate has no significant impact on the reaction heat. The regression equation with a reaction order of 0.9 can better describe the thermal denaturation process of PSC. Under constant temperatures of 34, 35, 36, 37, 38¡ãC and 39¡ãC, the D90 value of PSC denaturation (90% protein denaturation) The required time) are 53.76, 26.11, 15.75, 4.89, 4.26, 2.55?min respectively, the Z90 value (the temperature change at which the D value decreases by 90%) is 3.69?¡æ, and the apparent activation energy is 481.90?kJ/mol. The research results can provide theoretical reference for the further development and utilization of jellyfish collagen.
