Combined Application Of Stromal Cell-Derived Factor 1, Simvastatin And Collagen Scaffold For Ectopic Osteogenesis In Vivo

Abstract
Purpose: To explore the use of mouse stromal cell-derived factor 1 (msdf-1) combined with simvastatin (sim) and collagen scaffold (bio-oss?) to construct tissue-engineered bone without external seed cells. feasibility, and to test its effect on ectopic osteogenesis in vivo. Methods: 32 icr mice were randomly divided into 4 groups, with 8 mice in each group. Skin incisions were made on the skulls of mice, and mice in each group were implanted with: (1) 1:50 (volume ratio) dimethyl Sulfoxide (dimethylsulfoxide, DMSO)/phosphate buffered saline (PBS) mixture + collagen scaffold (blank control group); (2) 10-3mol/lsim solution + collagen scaffold (SIM group); ( 3) 200mg/lmsdf-1 solution + collagen scaffold (msdf-1 group); (4) 10-3mol/lsim + 200mg/lmsdf-1 solution + collagen scaffold (sim+msdf-1 group). One week after implantation, 50 ¦Ìl of the corresponding solutions of each group above were locally injected into the stent for 2 consecutive days every day. After 6 weeks of feeding, the scaffold and surrounding tissues were removed, and the osteogenic effect was qualitatively and quantitatively observed through soft X-ray projection imaging, gray scale measurement, He staining, and immunohistochemical staining. Results: The soft , p<0.01] and msdf-1 group [(255529.5¡À152142.4) pixels, p<0.05]; obvious expression of osteopontin and osteocalcin can be seen in the sim+msdf-1 group; sim+msdf-1 group The density of vascular plexus [(46¡À8) strips/mm2] was significantly higher than that of the blank control group [(23¡À7) strips/mm2, p<0.01] and sim group [(24¡À6) strips/mm2, p<0.01] . Conclusion: Tissue-engineered bone without external seed cells constructed using msdf-1 combined with SIM and collagen scaffold can ectopically form bone in the mouse skull subcutaneously.
