Study On Selecting Different Extractants To Prepare Channel Catfish Skin Collagen

Abstract
Different extraction agents were selected to prepare channel catfish skin collagen, and some qualitative studies were conducted on the collagen. Experimental results show that under 4¡ãC conditions, catfish skin is soaked in 0.1 mol/l naoh for 6 hours and then in 2.5% nacl for 6 hours. It can remove impurity proteins better than using naoh and nacl alone or at the same time. Using 10% isopropyl Propanol solution removes fat, and leaching with 0.1mol/l citric acid for 3 days can better extract collagen, which is colorless and odorless, and the extraction rate can reach 34.67%. The obtained collagen is rich in glycine, with a mass fraction of 21.19%, and its denaturation temperature is 50.2¡ãC. The sds-page experimental results show that the extracted catfish skin acid-soluble collagen has high purity, contains two ¦Á chains, and has a higher ¦Â chain content. Fish skin collagen solution has high viscosity, which increases with the increase of protein concentration.
