Abstract
Using fresh pig skin as raw material, Alcalase was used to hydrolyze collagen, and various factors affecting the Alcalase hydrolysis process were studied. By measuring the degree of hydrolysis and superoxide anion radical (O2-¡¤) scavenging rate, the Alcalase hydrolyzed pig skin was determined. Optimal conditions for collagen; study the scavenging effects of DPPH free radicals and O2-¡¤ on pig skin collagen hydrolyzate of different mass concentrations prepared under optimal conditions. The results show that the optimal conditions for Alcalase to hydrolyze pig skin collagen are: pH 7.5, temperature 55¡ãC, enzyme to substrate ratio 6000U/g, substrate mass concentration 40mg/mL, and hydrolysis time 4h; under these hydrolysis conditions, The degree of hydrolysis reaches 9.55%, and the O2- ¡¤ scavenging rate reaches 60.11%; within the corresponding mass concentration range of 10 to 50 mg/mL, the maximum scavenging rate of DPPH free radicals of the pig skin collagen hydrolyzate is 95.06%, and the IC50 is 3.89 mg/mL. ;O2-¡¤Maximum clearance rate is 65.89%, IC50 is 16.43mg/mL.
