Abstract
[] Objective To explore the effect of Smad7 gene on the expression of type I and type III collagen (ColI and ColIII) in rat glomerular mesangial cells (MsC), and to provide experimental basis for trying to use Smad7 for gene therapy in renal fibrosis. Methods: The recombinant expression plasmid containing Smad7 was transfected into rat MsC through liposome mediation, and G418 screening and Northernblot and Westernblot methods were used to identify the expression changes of transfected positive MsC clones Col¢ñ and Col¢ó using RT-PCR and Westernblot methods respectively. . Results The positive MsC clones (S-22 and S-26) that stably and highly expressed Smad7 were successfully established, and it was confirmed that the expression of Col¢ñ and Col¢ómRNA and protein of the two positive MsC clones were significantly inhibited, and the expression of Col¢ñ and Col¢ómRNA of S-22 clone was reduced respectively. 47% and 56%, and its protein expression decreased by 65% and 54% respectively. Conclusion Smad7 may reduce the progression of renal fibrosis by inhibiting the production of Col¢ñ and Col¢ó in tissues.
