Abstract
Objective To study the effect of infrared rays on the expression of c-Jun and type ¢ñ and type ¢ó collagen in fibroblasts (HSF) cultured in vitro, and to provide a theoretical basis for the molecular mechanism of photoaging damage caused by infrared rays. Methods: Primary skin fibroblasts were extracted and cultured. The fibroblasts were divided into control group (without infrared irradiation) and experimental group (infrared irradiation). MTT was used to detect cell activity in each group. Real-time quantitative PCR and immunocytochemistry (ICC) were used. Methods The expressions of c-Jun and type ¢ñ and ¢ó collagen mRNA and protein in each group were detected. Results MTT test showed that compared with the control group, infrared irradiation in each group of the experimental group had varying degrees of inhibitory effects on the proliferation of HSF; infrared irradiation down-regulated the expression of type ¢ñ collagen mRNA and protein. As the irradiation dose increased, the expression of type ¢ñ collagen decreased. The expression of mRNA and protein decreased more obviously (P<0.01); after 12 hours of infrared irradiation of fibroblasts, the expression of type III collagen mRNA and protein showed a dose-dependent decrease (P<0.05, P<0.01). The expression of type collagen mRNA and protein was up-regulated in a dose-dependent manner (P<0.05, P<0.01); infrared irradiation up-regulated the expression levels of c-Jun mRNA and protein in a dose-dependent manner as the irradiation dose increased. (P<0.05, P<0.01). Conclusion Infrared irradiation of human skin fibroblasts increases the expression of c-Jun, inhibits the expression of type ¢ñ collagen, and interferes with the expression of type ¢ó collagen.
