Abstract
Using the hydrolysis degree as an indicator, the conditions for the enzymatic hydrolysis of chicken breast cartilage type II collagen by neutral protease were optimized. On this basis, type II collagen enzymatic hydrolysis complexes with different hydrolysis degrees were prepared, and their immune activity was evaluated using the lymphocyte proliferation rate as an indicator. Research. The results show that neutral protease has the strongest enzymatic hydrolysis ability, and its optimal enzymatic hydrolysis conditions are: enzymatic hydrolysis temperature 50 ¡æ, pH 7.5, substrate mass concentration 25 mg/mL, enzyme addition amount 40 U/mg, enzymatic hydrolysis time 150 minutes. When the hydrolysis degree of type II collagen is 18%, its proliferation activity on lymphocytes reaches 58.69%. The mass concentration of type II collagen enzymatic complex also has a significant impact on the proliferation rate of lymphocytes. When its mass concentration reaches 1 mg/mL, the lymphocyte proliferation ability reaches the maximum. The molecular weight of type II collagen enzymatic complex with high immune activity is mainly distributed in the range of 1 000 to 180 D, accounting for 75.21% of its total content.
