Abstract
Objective: To study the physical and chemical properties of blue shark cartilage type II collagen isolated by alkali treatment-peptic hydrolysis method. Methods: Blue shark cartilage was freeze-dried and crushed, followed by alkali treatment, pepsin-limited hydrolysis, salting out and dialysis to obtain type II collagen. The extracted type II collagen was identified using SDS-PAGE, amino acid composition analysis, ultraviolet spectrum scanning and differential scanning calorimetry. Results: Blue shark type II collagen is composed of ¦Á1 chain with a molecular mass of 130kD. Glycine is the amino acid with the highest content of 356¡ë in blue shark type II collagen. It is rich in alanine, proline and hydroxyproline, and The amino acid residues of the peptide chain have the structural characteristics of Gly-XY. Blue shark type II collagen has an ultraviolet absorption peak at a wavelength of 226 nm, and the denaturation temperature is 41¡ãC. Conclusion: Blue shark type II collagen was obtained by alkali treatment and restriction enzyme hydrolysis, which has the typical characteristics of type II collagen.
