In Vitro Study Of New Plga/Collagen Composite Materials For Cartilage Regeneration

Abstract

Objective To design and prepare new lactic-co-glycolic acid copolymer [poly(lactic-co-glycolicacid), PLGA]/collagen (collagen) composite materials, study its promoting effect on cartilage regeneration in vitro, and provide new scaffold materials for cartilage tissue engineering. . Methods The collagen porous sponge was composited with PLGA woven omentum using freeze-drying technology; the surface microstructure of the material was observed with scanning electron microscope, and the pore size was counted using image software; bovine articular chondrocytes (BAC) were isolated and cultured, and inoculated into PLGA/glue Raw materials were used to detect cell inoculation efficiency, and scanning electron microscopy was used to observe cell growth inside the material; after 1 week of in vitro culture, DNA and glycosaminoglycan (GAG) content were detected, and real-time PCR was used to detect type I collagen, type II collagen, and aggregated protein. Aggrecan mRNA expression intensity. Bovine knee articular cartilage tissue and BACs cultured in monolayer in vitro were used as controls. Results: A new PLGA/collagen composite material was successfully constructed, with a surface pore size of (136.4¡À11.8) ¦Ìm; cell seeding efficiency of 87.8%¡À1.6%; BACs grew actively on the surface and center of the material. The DNA and GAG content after 1 week of culture, ¢ò The expression intensity of collagen and aggrecan mRNA was significantly higher than that of the control group (P<0.05). Conclusion The newly designed PLGA/collagen composite material can promote cartilage regeneration in vitro and can be used as a scaffold material for cartilage tissue engineering research.
