Abstract
In order to improve the production of human-like collagen by high-density fermentation of recombinant Escherichia coli BL21, the effects of induction timing, acetic acid concentration, induction intensity and nitrogen supplementation methods on the expression of exogenous genes were studied, and each factor was optimized. Fed-batch fermentation was adopted, and the initial liquid volume was 6L. The final cell density can reach 84.3g/L, and the expression level of human-like collagen is 14.6g/L. The results show that induction is carried out in the middle and late stages of logarithmic growth to minimize the accumulation of acetic acid. After 3 hours of induction at 42¡ãC, the temperature is lowered to 39¡ãC to continue induction. A periodic operation mode of adding 36 ml of feed nitrogen solution every 10 minutes is beneficial.
