Abstract
Collagen-chitosan composite scaffolds were prepared by freeze-drying and structurally modified with laminin (LN). Their physical properties were measured, and a three-dimensional culture system of fetal rat hippocampal neural stem cells (NSCs) was established. The NSCs were also investigated. Biocompatibility with collagen-chitosan composite scaffold. Comparison of parameters such as pore size, porosity, water absorption, and degradation rate showed that the composite scaffold with a volume ratio of 7:3 was more suitable for the requirements of in vitro cell culture, and the seeding rate of NSCs on the LN-modified scaffold material was significantly improved. Laser confocal microscopy and scanning electron microscopy observations showed that NSCs can grow, proliferate and differentiate well within the collagen-chitosan composite scaffold. These results lay a solid experimental foundation for further clinical transplantation applications of NSCs, as well as new drug screening and mechanism research for the treatment of neurological diseases.
