Effect Of Integrin Beta On The Adhesion Between Liver Cancer Cells And Type Iv Collagen

Abstract
Using micropipette experimental technology, we studied the treatment of hepatocellular carcinoma (HCC) cells with a monoclonal antibody (Anti-CD29) against the integrin beta1 subunit (CD29), and observed the effect of Anti-CD29 on the cells and type IV collagen-coated surfaces. The effect of adhesion was analyzed by flow cytometry on the expression of integrin beta1 subunit on the surface of HCC cells. The results showed that the adhesion force between HCC cells and the surface lined with 5 ¦Ìg/mL type IV collagen was 932¡À134 (¡Á10-10N, n=60). The adhesion force decreased to 449¡À119 after adding 5 ¦Ìg/mL Anti-CD29. (¡Á10-10N, n=60); the adhesion force decreases to 220¡À78 when 10 ¦Ìg/mL Anti-CD29 is added (¡Á10-10N, n=55); the adhesion force decreases when 20 ¦Ìg/mL Anti-CD29 is added to 222¡À67 (¡Á10-10N, n=65). Flow cytometry analysis showed that the expression rate of integrin beta1 subunit in the cells studied reached 95.78%. The above results indicate that integrin beta1 subunit is an important receptor basis for the adhesion of HCC cells to type IV collagen-coated surfaces.
