Abstract
Objective To explore the changes in extracellular matrix proteins and collagen in aortic vascular remodeling in spontaneously hypertensive rats (SHR) and to analyze the effects of Xuezhikang intervention. Methods 30 8-week-old male SHR rats were randomly divided into Xuezhikang low-dose group [20 mg/(kg¡¤d), n10], Xuezhikang high-dose group [200 mg/(kg¡¤d), n10] and placebo. dosage group (n10), and male Wistar rats of the same age served as the control group (n10). All rats were fed until they were 16 weeks old, venous blood was drawn and centrifuged, and the serum was separated and the enzyme-linked immunosorbent assay was used to determine serum matrix metalloproteinase 9 ( MMP-9) and tissue-type matrix metalloproteinase inhibitor (TIMP-1) levels; at the same time, the thoracic aorta was taken, and HE staining was used to analyze the blood vessel wall thickness and wall/lumen ratio; Masson’s trichrome staining was used to analyze changes in vascular collagen fibers; immune tissue Chemical staining was used to analyze the protein expression of MMP-9 and TIMP-1 in the thoracic aorta, and Image-Pro Plus 6.0 software was used to perform grayscale quantification on immunohistochemistry stained sections. Results The serum MMP-9 level of 16-week-old rats in the placebo group was significantly higher than that in the control group (124.60¡À30.01 ¦Ìg/L vs. 66.00¡À22.67 ¦Ìg/L, P<0.01); after treatment with Xuezhikang, high-dose Xuezhikang The serum MMP-9 level of the treatment group was significantly lower than that of the placebo group (67.96¡À8.80 ¦Ìg/L vs. 124.60¡À30.01 ¦Ìg/L, P<0.05). Serum TIMP-1 levels in SHR rats were significantly higher than those in the control group, and the difference was statistically significant (P<0.01); however, there was no statistically significant difference between groups in hypertensive rats (P>0.05). Compared with the thoracic aorta of 16-week-old SHR rats (placebo group) and Wistar rats (control group), it can be seen that the blood vessel wall is significantly thickened, the wall/lumen ratio is increased (P<0.05), and the elastic fibers are arranged disorderly. The collagen component of the tube wall increased significantly; after Xuezhikang treatment, the tube wall/lumen ratio of the Xuezhikang high-dose group was significantly lower than that of the placebo group (6.8%¡À0.9% vs. 7.8%¡À0.7%, P<0.05). Collagen decreased in all groups, and the amount of collagen in the Xuezhikang high-dose group was close to that of the control group. Immunohistochemical staining of the thoracic aorta showed that the expression of MMP-9 in the placebo group was significantly stronger than that in the control group and Xuezhikang treatment group; after grayscale quantification, the grayscale value of MMP-9 in the placebo group was significantly higher than that in the Xuezhikang high-dose group and Xuezhikang treatment group. In the control group, the difference was statistically significant (P<0.01). The expression of TIMP-1 was not obvious in each group, and the quantitative difference in gray scale was not statistically significant. Conclusion Hypertensive rats have vascular remodeling, which is manifested by increased MMP-9 levels and increased collagen deposition on the aortic wall. Xuezhikang can inhibit MMP-9 levels and collagen deposition and reduce the degree of vascular remodeling.
