Abstract
This article studied the feasibility of using subtilis alkaline protease (Alcalase 2.4L) and Aspergillus niger acid protease (3000U/g) to synergistically hydrolyze pig skin collagen to prepare oligopeptide hydrolysates under gradient pH conditions without the addition of alkali. sex. The effects of single-enzyme single-factor hydrolysis conditions and double-enzyme adding methods on the pig skin degradation rate and protein hydrolysis degree were investigated. On this basis, the best method for hydrolyzing pig skin under the one-time double-enzyme feeding scheme was further optimized through orthogonal experiments. Conditions: The hydrolysis temperature is 60¡ãC, the alkaline protease addition amount is 25 ¦Ìl/g protein, the acid protease addition amount is 4% (g/g), the substrate concentration is 60g/L, and the hydrolysis time is 12 hours. Under the above-mentioned optimal conditions, the degradation rate of fresh pig skin can reach 94.7%, the degree of protein hydrolysis can reach 17.7%, and the selectivity of oligopeptides with a molecular weight less than 1000 (peptides below 10 amino acids) in the hydrolyzate reaches 42.5%. . The results show that without adding alkali, the dual-enzyme synergistic hydrolysis method can significantly improve the pig skin degradation rate, especially the protein hydrolysis degree.
