Preparation Of Collagen Sponge Loaded With Bfgf-Plga Microspheres And Study On Its Sustained Release Properties In Vitro

Abstract
The introduction of growth factors with controlled release behavior into scaffold materials aims to structurally and functionally simulate the energy of extracellular matrix. A collagen sponge sustained-release system loaded with bfgf-plga microspheres was prepared to explore its release performance of bfgf in vitro. The double emulsion solvent evaporation method was used to make microspheres, and the microspheres were composited with collagen sponges; the surface morphology and structure of the microspheres and the collagen sponge loaded with microspheres were observed through scanning electron microscopy; the drug loading and encapsulation of the drugs in the microspheres were tested using the ELISA method rate, and the in vitro release behavior of bfgf in microspheres and microsphere-loaded collagen sponges was studied. The results show that the surface of the microspheres is smooth, the drug loading capacity is 0.0599%¡À0.0019%, and the encapsulation rate is 79.9%¡À2.8%; the burst release rate of the sponge loaded with microspheres is 14.6%, which is lower than the 20.0% level of the microspheres themselves; Moreover, the in vitro release time of bfgf from the microsphere-loaded sponge is longer than that of the microsphere itself. The collagen sponge sustained-release system loaded with bfgf-plga microspheres can stably release bfgf in vitro, with a small burst release rate, and is expected to develop into an ideal tissue engineering scaffold material.
