Abstract
new injectable thermosensitive chitosan-glycerophosphate-collagen (C-GP-Co) hydrogel was prepared and its interaction with adipose tissue-derived stem cells (ADSCs) was tested. ) biocompatibility. First, mix a chitosan solution with a volume fraction of 2.2% and a ¦Â-sodium glycerophosphate solution with a mass fraction of 50% in a certain proportion, then add 2 mg/mL collagen solution to the above solution and mix to obtain chitosan-sodium glycerophosphate-collagen. The hydrogel was then used to observe its microstructure with a scanning electron microscope, the gelation time at 37¡ãC was measured and the osmotic pressure of the culture medium extracted from the hydrogel was measured; finally, the ADSCs that were isolated, cultured and immunophenotyped were inoculated in Culture in C-GP-Co hydrogel, observe the growth morphology of cells under an inverted microscope, and use Live/DeadViability-Cytotoxicity kit to stain and observe cell apoptosis, and CCK-8 kit to measure cell proliferation. The results show that the gelation time of C-GP-Co hydrogel at 37¡ãC is 12 minutes, the interior of the hydrogel has a sponge-like porous structure, and the osmotic pressure of the extraction medium in the hydrogel is 310~330mmol/kg. After 7 days of culture, the ADSCs on the surface of the C-GP-Co hydrogel adhered and grew and showed good cell activity. The above results indicate that the prepared C-GP-Co hydrogel has good cytocompatibility and can be used as a good scaffold for the growth and proliferation of ADSCs.
