Abstract
Using the skirt of Chinese soft-shell turtle as raw material, a single factor experiment was used to explore the effects of enzyme addition, ultrasonic time, ultrasonic power, and material-liquid ratio on collagen. A mathematical model was established through the Box-Behnken response surface method to obtain the optimal enzyme extraction process. , to compare the thermal stability performance of conventional enzymatic extraction and ultrasound-assisted enzymatic extraction of collagen. The results show that when the extraction solvent is acetic acid (0.5?mol/L) and the solid-liquid ratio is 1:20 (g/mL), the optimal process parameters for collagen extraction using ultrasonic enzyme are 0.8% enzyme addition, ultrasonic The time was 43?min and the ultrasonic power was 176?W. Under these conditions, the collagen yield was 74.50%. The results of the differential scanning calorimetry experiment showed that the thermal stability of ultrasonic enzymatically extracted collagen was significantly better than that of conventional enzymatically extracted collagen. This research can provide a certain scientific basis for the deep processing and application of Chinese soft-shell turtle skirt collagen.
