Abstract
Abstract: Objective To explore the effect of Bushen Huoxue Recipe on rat fibroblast proliferation and collagen expression, and to provide experimental basis for its anti-myocardial fibrosis. Methods The medicated serum of Bushen Huoxue Recipe was prepared through serum pharmacology method, and then mixed with DMEM to form cell culture medium. Rat cardiac fibroblasts (cfs) were extracted and subcultured. The 3rd to 4th generation cfs were divided into 4 groups: group a was cultured in 10% medicated serum culture medium, group b was cultured in 20% medicated serum culture medium, and group c was cultured in 20% medicated serum culture medium. Group d was cultured with drug-free serum culture medium, and group d was the control group, cultured with conventional fetal bovine serum culture medium. After 72 hours of culture, cck-8 was used to detect cfs proliferation, rt-PCR was used to detect collagen gene expression, and western blotting was used to detect collagen expression. Results Compared with groups c and d, the cfs proliferation in groups a and b was significantly reduced, and group b was further reduced compared with group a (p<0.05); the expression of type I and type III collagen mRNA in groups a and b was lower than that in group c and group b. d was significantly down-regulated in two groups (p<0.05), and the expression of type ¢¡ and ¢£ collagen in groups a and b was significantly lower than that in groups c and d (p<0.05). Conclusion Bushen Huoxue Recipe can inhibit the proliferation and collagen expression of rat fibroblasts.
