Abstract
In order to explore a rapid determination method for the content of collagen peptides derived from tilapia, tilapia skin collagen peptides were used as reference substances to improve the biuret colorimetric method for determining the peptide content, and the maximum absorption wavelength of the color complex was investigated. , peptides with high linear fitting degree and trichloroacetic acid (TCA) dosage range and pH value; repeatability, precision evaluation and sample recovery experiments of the standard curve were carried out under optimized conditions; the effect of this method on different Applicability of molecular weight tilapia skin and fish scale collagen peptide system detection. The results show that when the mass concentration of collagen peptide (nominal molecular mass 3 kDa) is in the range of 0.3 to 1.5 mg/mL, use 13% TCA solution, adjust the system pH to 12.5, and detect at a wavelength of 545 nm. The absorbance is related to the peptide mass concentration. The linear fit is good (R2=0.999), and the standard curve equation is y=0.144 9x+0.001 5; the relative standard deviation (RSD) obtained by the repeatability and precision experiments is 1.86% and 1.84% respectively. Sample addition and recovery The rates are 113.9% and 109.7% respectively, and the RSDs are 1.39% and 1.00% respectively; the linear range is wide, the repeatability is good, and the accuracy is high. This method was applied to the detection of different types of tilapia peptide product systems. The sample recovery rates were similar and the deviations were small, indicating that this method is simple, fast and suitable for the detection of tilapia-derived peptide content.
