Abstract
In order to explore the effect of cell matrix type I collagen (col-¢¡) on the metastasis and proliferation ability of head and neck squamous cell carcinoma cells, head and neck squamous cell carcinoma cell lines were used as the object. The transwell chamber was used to examine the in vitro migration ability of the cells, and the inverted microscope imaging system was used to detect the cells. The movement speed and diffusion ability, western blotting or/and immune cell fluorescence staining were used to detect the expression of the cell surface adhesion molecule e-cadherin and the cellular localization of ¦Â-catenin, and the expression levels of mtt and pcna were used to evaluate cell proliferation. The results showed that col-¢¡ can promote the migration of head and neck squamous cell carcinoma cells, increase the speed of cell movement, and accelerate cell spread. The possible mechanism is by up-regulating the phosphorylation of ¦Â-catenin, thereby down-regulating the expression of the adhesion protein e-cadherin. Col-¢¡ can also promote the proliferation of head and neck squamous cell carcinoma cells. The possible mechanism is to increase the nuclear translocation of ¦Â-catenin and increase the expression of cyclin d1 (cyclind1). Research results show that col-¢¡ enhances the metastasis and proliferation capabilities of head and neck squamous cell carcinoma cells by upregulating ¦Â-catenin phosphorylation levels and promoting ¦Â-catenin nuclear translocation.
