Abstract
Objective To explore the effect of t-2 toxin on the synthesis of type II collagen and proteoglycan, the main interstitial components of chondrocytes, and the protective effect of selenium. Methods: MTT method was used to detect the effect of T-2 toxin on the survival rate of chondrocytes; TB staining was used to detect proteoglycan expression in human chondrocytes; immunohistochemistry was used to detect collagen ¢¢ expression in human chondrocytes; rt-PCR was used to detect human chondrocyte ¢¢ Collagen and proteoglycan mRNA expression. Results The effect of t-2 toxin on chondrocytes showed a typical concentration-dependent and time-dependent relationship within the concentration range of 0.001-2mg/l, and with the extension of the action time, the concentration-dependent relationship became more obvious. T-2 toxin (10-20 ¦Ìg/l) can inhibit the synthesis of type II collagen and proteoglycan and its mRNA expression in chondrocytes. Adding selenium can significantly stimulate the expression of proteoglycan mRNA in chondrocytes, partially weaken the inhibition of T-2 toxin, and have a certain protective effect, but it does not show the effect on the expression of type II collagen and its mRNA caused by T-2 toxin. protective effect. Conclusion T-2 toxin has a significant inhibitory effect on the synthesis of type II collagen and proteoglycans in chondrocytes, and the trace element selenium has a certain protective effect on the changes in chondrocyte proteoglycans caused by t-2 toxin. ?More restores
