Abstract
Extracting insoluble collagen (ISC) from human and animal bones unearthed from archaeological sites and conducting C and N stable isotope analysis is currently the main research method for ancient recipe analysis. However, for bone samples from which ISC cannot be extracted, reconstruction of ancient recipes is impossible. Previously, we successfully extracted soluble collagen (SC), the degradation product of ISC in ancient bones, for the first time through gel chromatography, and found that SC and ISC have similar C and N stable isotope ratios and can also be used in ancient bone. Recipe research. In order to further explore the mechanism of similar stable isotopes between the two, this paper selected more ancient bone samples and carried out analysis of element content, stable isotope values and amino acid composition of SC and ISC. Research results show that the total amino acid composition of SC and ISC is completely consistent with the characteristics of type I collagen. In addition, compared with ISC, the composition of 8 essential amino acids in SC is very close, with a variation range of only 0.25% (n=3); while the variation in 11 semi-essential amino acids and non-essential amino acids is slightly larger (0.90%, n= 3). Obviously, the slight difference in the content of amino acids of different properties is the main reason why the C and N stable isotopes of the two proteins are similar but not the same.
