Relationship Between Human Leukocyte Antigen-Dr4/1 Phenotype, Type Ii Collagen-Specific T Cell Proliferation And Serum Type Ii Collagen Antibodies In Patients With Rheumatoid Arthritis

Abstract
Objective: To explore the relationship between hla-dr4/1 phenotype, type II collagen (c¢¢) 263-272 polypeptide-specific T cell proliferation and serum c¢¢263-272 antibodies in patients with rheumatoid arthritis (RA). Methods: c¢¢263- The 272 polypeptide was incubated with peripheral blood mononuclear cells (PBMC) of 70 RA patients for 5 days. T cell proliferation was measured using the 3h entry method, and the patient’s serum c¢¢263-272 antibody level was detected using the enzyme-linked immunosorbent assay (ELISA). Based on the sequence Specific primer polymerase chain reaction (PCR-SSP) technology was used to perform hla-dr typing on the above patients. Results: Among 70 RA patients, 27 were hla-dr4/1 phenotypic positive and 43 were hla-dr4/1 The phenotype is negative. Among RA patients with a positive hla-dr4/1 phenotype, c¢¢263-272 polypeptide can stimulate T cell proliferation in 15 cases (55.6%), and 12 cases (44.4%) showed no response; 18 cases ( 66.7%) patients were positive for anti-c¢¢263-272 antibodies. Among RA patients with negative hla-dr4/1 phenotype, 13 cases (30.3%) of T cells were reactive to c¢¢263-272 polypeptide, and 30 cases were non-reactive. (69.7%); 15 RA patients (34.8%) were positive for anti-c¢¢263-272 antibodies. The c¢¢263-272-specific cell proliferation and c¢¢263-272 antibody production in the hla-dr4/1 phenotype-positive group were higher than those of hla-dr4/1 Phenotype negative group. Conclusion: The proliferation of c¢¢263-272-specific T cells and the production of c¢¢263-272 antibodies in RA patients are related to the HLA-DR4/1 phenotype.
