Effects Of Paeoniflorin On Glucocorticoid Receptors In Peripheral Blood Mononuclear Cells Of Rats With Collagen-Induced Arthritis

Abstract
To study the effect of paeoniflorin (PF) on the glucocorticoid receptor (GCR) of peripheral blood mononuclear cells (PBMCs) in rats with collagen-induced arthritis. The collagen-induced arthritis (CIA) rat model was used. On the 14th day after the initial immunization, PF25, 50, 100mg¡¤kg-1 or triptolide 20¦Ìg¡¤kg-1 or PF50mg¡¤kg-1+RU48615mg¡¤ were administered intragastrically. kg-1, once a day, administered continuously for 28 days. On the 2nd day after the last dose, all experimental rats except the PF+RU486 group had their eyeballs removed to collect blood. They were sacrificed and unimmunized joints were collected from all rats. The expression of GCR¦Á and GCR¦Â in PBMCs was detected by ELISA and reverse transcription-polymerase chain reaction. The expression of GCR¦Á and GCR¦Â mRNA in PBMCs was analyzed by (RT-PCR) method, and the expression of IL-1¦Â, NF-¦ÊBp65, TNF-¦Á, and PGE2 in joint synovial tissue was analyzed by immunohistochemistry. PF50, 100mg¡¤kg-1 can inhibit the expression of IL-1¦Â, NF-¦ÊBp65, TNF-¦Á, and PGE2 in the synovial tissue of CIA rats (P<0.05, P<0.01), while RU48615mg¡¤kg-1 can significantly affect This effect of PF50mg¡¤kg-1 (P<0.05); PF50 and 100mg¡¤kg-1 can upregulate the expression of GCR¦Á and GCR¦ÁmRNA, and downregulate the expression of GCR¦Â and GCR¦ÂmRNA (P<0.01). PF inhibits the joint synovium of CIA rats. Inflammatory mediators may be related to their regulation of GCR expression.
