Abstract
Objective To observe the effect of Wenhua Jubi Recipe on the expression of DNA methyltransferases (DNMTs) in peripheral blood mononuclear cells (PBMCs) of rats with collagen-inducing arthritis (CIA). Effect, to explore the mechanism of Wenhua Juanbi Recipe in treating CIA. Methods: 90 Wistar rats were randomly divided into a modeling group (80 rats) and a normal control group (10 rats, referred to as the normal group). In the modeling group, bovine type II collagen emulsion was injected into the tail root of rats to prepare the CIA model. The 50 rats that were successfully modeled were randomly divided into model group, methotrexate group (MTX), and Wenhua Juanbi Recipe low, medium, and high-dose groups (referred to as traditional Chinese medicine low-, medium-, and high-dose groups). 10. The model group was given normal saline by gavage; the low, medium and high dose groups of traditional Chinese medicine were given Wenhua Juanbi Recipe 22.9, 45.8 and 68.7 g/(kg¡¤d) by gavage respectively, once a day; the MTX group was given 0.78 mg/ MTX suspension was administered orally at a dose of kg once a week for 30 consecutive days. Toe swelling was assessed using volumetric method (drainage volume). After drug intervention, PBMCs from rats in each group were extracted, and real-time quantitative PCR was used to detect the mRNA expression levels of DNMTs (DNMT1, DNMT3a and DNMT3b) in PBMCs. Results Compared with the normal group, the toes of the rats in the model group were significantly swollen (P<0.01); compared with the model group, the toe swelling of the rats in the low-, medium-, and high-dose traditional Chinese medicine groups and the MTX group was significantly reduced (P<0.01). Compared with this group before treatment, the toe swelling of rats in each dose group of traditional Chinese medicine and the MTX group was reduced (P<0.01). Compared with the normal group, the expression levels of DNMT1, DNMT3a and DNMT3b in rat PBMCs in the model group were significantly reduced (P<0.01); compared with the model group, the expression levels of DNMT1, DNMT3a and DNMT3b in each dose group of traditional Chinese medicine and the MTX group were significantly increased (all P<0.01). 0.01); there was no statistically significant difference in the expression levels of DNMT1, DNMT3a and DNMT3b among the various dosage groups of traditional Chinese medicine (P>0.05). Conclusion The expression of DNMTs in PBMCs of CIA rats is reduced. Wenhua Jubi Recipe can increase the expression of DNMTs in PBMCs of CIA rats without obvious dose dependence. By upregulating the expression of DNMTs, regulating the methylation status of CIA rats may be one of its mechanisms for treating CIA.
