Hydrolysis Process Of Fish Scale And Its Effect On Properties Of Collagen Peptide

Abstract

Using grass carp (Ctenopharyngodon idellus) scales as raw materials, the effect of the hydrolysis process on the properties of enzymatically prepared fish scale collagen peptides under the action of three commonly used industrial proteases was studied. The results showed that: for alkaline protease, after hydrolysis for 4 hours under the test conditions, the degree of hydrolysis was 9.5%. At this time, the comprehensive performance of collagen peptide was the most ideal, the clearance rate of DPPH? was 82.14%, and the emulsification activity index was 70.12 m2 /g. For the complex protease, after hydrolysis for 4 hours under experimental conditions, the degree of hydrolysis was 8.7%, the clearance rate of DPPH? by collagen peptide was 86.32%, and the emulsification activity index was 32.93 m2/g. For neutral protease, after hydrolysis for 4 hours under experimental conditions, the degree of hydrolysis was 7.2%, the clearance rate of DPPH? by collagen peptide was 70.15%, and the emulsification activity index was 36.88 m2/g. The above results show that there are differences in the properties of collagen peptides with different degrees of hydrolysis. Different proteases are used for hydrolysis. When the degree of hydrolysis is the same, the properties of the obtained collagen peptides are also different. Three kinds of industrial proteases were applied to investigate the effect of hydrolysis process on the functional properties of crass carp(Ctenopharyngodon idellus) scale derived collagen peptide.The results revealed that collagen peptide with different DH and the same DH by different proteases showed different properties. For alcalase 2.4L FG group,after 4 h of hydrolysis,the clearance of DPPH free redical,emulsifying activity index (EAI) and DH were 82.14%,70.12 m2/g and 9.5%,respectively.For protamex group and neutrase 1.5 MG group , after 4 h of hydrolysis, the DH of collagen peptide were 8.7% and 7.2%, the clearance of DPPH free redical were 86.32% and 70.15%, and the EAI were 32.93 m2/g and 36.88 m2/g, respectively.
