Abstract
A method for isolating and preliminarily identifying rat sinoatrial node pacemaker cells using collagenase is introduced. The operation is simple and convenient, and the amount of enzyme used is small. The isolated pacemaker cells have similar action potential characteristics to the in situ sinoatrial node pacemaker cells: there is obvious automatic depolarization during diastole, the average maximum diastolic potential is -55mV, the average action potential amplitude is 58mV, the average APD50 is 18ms, and the average APD90 29ms. Provide appropriate specimens for further electrophysiological and histochemical studies.
