Abstract
Abstract Purpose: To explore the effect of plasmid DNA-modified nano-¦Â-tricalcium phosphate/type I collagen solution composite containing human bone morphogenetic protein-2 as the target gene on the proliferation of mouse preosteoblasts. Methods: Nano-¦Â-TCP/type I collagen solution composite material was modified with plasmid DNA containing hBMP-2 as the target gene, and an in vitro culture system of MC3T3-E1 cell line and nanocomposite material was established. They were divided into scaffold groups and dish groups, and the cytological morphology was observed with scanning electron microscope and light microscope. Cell proliferation was detected by Alamar Blue method, cell cycle was detected by MuseTM Cell Analyzer method, and cell proliferation genes such as Cdk2 and Cdk4 were detected by real-time fluorescence quantitative PCR. Results: The cell adhesion number, proliferation number and surface distribution of the composite material were higher than those of the plate group (P<0.05). The experimental results showed that the scaffold group was better than the plate group. Conclusion: Compared with plasmid DNA simply containing hBMP-2 as the target gene, the composite material can better promote the adhesion, proliferation and functional metabolism of preosteoblasts, indicating that it has good cytocompatibility with preosteoblasts and can become a A new type of bone repair or tissue engineering material with promising applications For further information of this article and research, feel free to contact our team for asssitance.
