Abstract
Objective To explore the effect of Miao medicine Jinwu Jiangu prescription on the expression of nuclear factor kappa B (NF-¦ÊB) and serum IL-17 in joint synovial cells of rats with collagen-induced arthritis (CIA) model. Influence. Methods 60 Wistar rats were randomly divided into blank control group, model group, Jinwu Jiangu Recipe high, medium and low dose groups and Tripterygium wilfordii polyglycoside group, with 10 rats in each group. Except for the blank control group, the other groups were established For the CIA rat model, intragastric administration was started on the 7th day of modeling. Rats were sacrificed after 4 weeks of administration. Serum was collected, joint synovium was separated, and serum IL-17 was detected using enzyme-linked immunosorbent assay (ELISA). level, Western blot was used to detect the expression levels of NF-¦ÊB/P65, NF-¦ÊB/P50 and I¦ÊB¦Á proteins in joint synovial tissue. Results Compared with the blank control group, the serum IL-17 level of rats in the model group increased (P<0.01); compared with the model group, the serum IL-17 expression levels of the Jinwu Jiangu Recipe high and medium dose and Tripterygium wilfordii polyglycoside groups were significantly higher. decreased (P<0.01). Western blot results showed that compared with the blank control group, the protein activities of NF-¦ÊB/P65, NF-¦ÊB/P50 and I¦ÊB¦Á in the joint synovium of rats in the model group were enhanced (P<0.01); compared with the model group, Jinwu Jiangu prescription had higher , the NF-¦ÊB/P65 and NF-¦ÊB/P50 protein activities in the middle-dose and Tripterygium wilfordii polyglycoside groups were significantly reduced (P<0.05, P<0.01). The above indicators in the Jinwu Jiangu prescription low-dose group were higher than those in the Tripterygium wilfordii polyglycosides group. group, the difference was statistically significant (P<0.05, P<0.01). Conclusion Miao medicine Jinwu Jiangu prescription can reduce the expression of serum IL-17 in CIA model rats and inhibit the protein activities of NF-¦ÊB/P65 and NF-¦ÊB/P50.
