Abstract
Objective: To explore the effect of glucosamine hydrochloride on the expression of type I collagen and osteocalcin in the subchondral bone of knee joints with knee osteoarthritis. Methods: 46 postmenopausal female patients who underwent artificial knee joint surface replacement due to knee osteoarthritis were selected. Among them, 23 patients were treated with glucosamine hydrochloride for 12 weeks within six months before surgery, which was the observation group, and 23 patients who did not take it were the control group. Group. During the operation, 0.5 cm3 of subchondral bone tissue after tibial plateau osteotomy was collected from the two groups of knees, and bone tissue micro-CT detection, type ¢ñ collagen picric acid-sirius red staining, osteocalcin immunohistochemical staining, and type ¢ñ collagen were performed respectively. Results: Micro-CT examination found that the relative bone volume, trabecular space, trabecular thickness, and number of trabecular bones in the observation group were improved compared with the control group, and type I collagen and bone in the observation group were improved. Conclusion: Glucosamine hydrochloride may improve the microstructure of the subchondral bone of the knee joint in menopausal patients with female knee osteoarthritis by regulating the expression of type I collagen and osteocalcin, thereby improving the function of the subchondral bone of the knee joint. Aim: To research the effect of glucosamine hydrochloride on type ¢ñcollagen and osteocalcin level in knee subchondral bone of patients with knee osteoarthritis.Methods: Forty-six postmenopausal female patients with knee osteoarthritis underwent TKA surgery were allocated into 2 groups, those accepted glucosamine hydrochloride oral administration for 12 weeks during the half year before surgery were observation group, and those without the glucosamine hydrochloride treatment were control group. The subchondral bone tissue(0.5 cm3) of tibial in the 2 groups was collected and detected by micro-CT, collagen picric acid sirius red staining, osteocalcin immunohistochemical staining, as well as collagen ¢ñ and osteocalcin Western blot detection. Results: Compared with control group, the relative bone volume, intertrabecular spaces, trabecular thickness and numbers of trabecular all improved in the observation group. According to collagen picric acid sirius red staining and immunohistochemical staining, the contents of type ¢ñ collagen and osteocalcin in the observation group were higher than those in control group (P£¼0.05). According to the results of Western blot, the expression levels of type ¢ñ collagen and osteocalcin in the observation group were higher than those in control group (P£¼0.05).Conclusion: Glucosamine hydrochloride can effectively improve the subchondral bone microstructure and function of knee in female patients with knee osteoarthritis by regulating the expressions of type ¢ñ collagen and osteocalcin in subchondral bone tissue For further information of this article and research, feel free to contact our team for asssitance.
