Study On The Relationship Between Endometrial Matrix Metalloproteinaseª²2,ª²9 Mrna Expression And Endometritis In Dairy Cows

Abstract
In order to explore the mechanism of matrix metalloproteinase (mmps)ª²2 and mmpª²9 in dairy cow endometritis, 10 healthy Chinese Holstein cows and 10 Chinese Holstein cows suffering from acute suppurative endometritis 6 to 10 days postpartum were selected, respectively. For the control group and the experimental group, the concentrations of pgf2a and progesterone were detected by ELISA; the expression of type I collagen and type IV collagen in the endometrium was detected by immunohistochemistry; the expression of mmpª²2 and mmpª²9 mRNA in the endometrium was detected by sybrgreeni fluorescence quantitative PCR. The results showed that: the pgf2a concentration in the test group was extremely significantly lower than the control group (p<0.01), the progesterone content was significantly higher than the control group (p<0.05), and there was a significant negative correlation between pgf2a and progesterone concentration (r=0.893); the test group The expression levels of type I collagen and type IV collagen in the endometrium were significantly higher than those in the control group (p<0.01); the levels of mmpª²2 and mmpª²9 mRNA were significantly lower than those in the control group (p<0.01). The results suggest that the low expression of mmpª²2 and mmpª²9 in the endometrium of cattle with postpartum endometritis disease leads to obstruction of the degradation of type I and type IV collagen, resulting in insufficient stimulation signals provided by the extracellular matrix for endometrial cells, and uterine secretion. The amount of pgf2a is small, the corpus luteum is regressed, and the serum progesterone content is high; indicating that the expression of mmpª²2 and mmpª²9 mRNA in the endometrium promotes the occurrence and development of postpartum endometritis.
