Growth Ability Of Human Dental Pulp Cells On Three Bioactive Scaffolds

Abstract
Purpose: To observe the proliferation and differentiation of human dental pulp cells (HDPCs) on bioactive scaffolds. Methods: HDPCs were cultured using enzyme digestion method and passed to the 4th generation for experiments. Immunohistochemistry was used to identify cells and determine the expression rate of stromal precursor cell antigen-1 (stromal precursorantigen-1, stro-1). Three types of scaffolds were used in the experiment, including collagen (collagen, col) scaffold, collagen-bioactive glass (collagen-bioglass, col-bg) scaffold and collagen-bioglass-polycaprolactone (collagen-bioglass-polycaprolactone, col- bg-pcl) stent. HDPCs were implanted into the scaffold, and HDPCs proliferation was measured using the MTT method at 6h, 1d, 3d, 5d, 7d, 14d and 21d, and alkaline phosphatase staining was performed on the 14th day. Results: The HDPCs used in the experiment contained stro-1 positive cells; the proliferation of HDPCs on col-bg scaffolds and col-bg-pcl scaffolds was significantly higher than that on col scaffolds (p<0.05), especially on 14d and 21d col-bg scaffolds and col -The optical density value of the bg-pcl scaffold is 5 times that of the col scaffold; the alkaline phosphatase staining area on the col-bg scaffold and col-bg-pcl scaffold is significantly wider than that of the col scaffold. Conclusion: HDPCs proliferate and differentiate actively on col-bg scaffolds and col-bg-pcl scaffolds, which are better than traditional col scaffolds.
