Study On The Effects Of Astragalus On The Proliferation And Collagen Secretion Of Hepatic Stellate Cells In Rats

Abstract
Objective: To observe the effect of Astragalus on the proliferation and collagen secretion of rat hepatic stellate cells. Methods: Rat hepatic stellate cells were isolated and cultured. After one generation, different concentrations of astragalus were added for 48 hours, and the effect on cell proliferation was detected by MTT colorimetry. After the first generation cells were treated with 2 mg/ml for 48 hours, the changes in the secretion of type ¢ñ, ¢ó and ¢ô collagen were detected by ELISA. Results: 0.75-50mg/ml Astragalus (filtered through 0.45?m filter membrane) can reduce the MTT conversion rate and inhibit cell proliferation in an obvious dose-dependent manner; 2mg/ml Astragalus can secrete type ¢ñ, ¢ó and ¢ô collagen Volume decreases. Conclusion: Part of the anti-fibrosis mechanism of Astragalus lies in inhibiting the proliferation of activated hepatic stellate cells and the synthesis of collagen, thereby reducing the deposition of collagen fibers in the liver.
