Abstract
Objective To observe the effect of Liuyu Tongzhi on the content of type I and type III collagen and the expression of matrix metalloproteinase-9 (MMP-9) in vulnerable plaques in mice with atherosclerosis, and to explore its mechanism of stabilizing vulnerable plaques. . Methods ApoE gene knockout mice were fed high-fat diet to establish a vulnerable plaque model. The mice were divided into a normal group, a model group, a high-dose traditional Chinese medicine group, a low-dose traditional Chinese medicine group, and a simvastatin group, with 10 mice in each group. The drug administration group was given corresponding drug intervention, and the model group and the normal group were given an equal amount of normal saline. After 12 weeks, picric acid-Sirius red staining was used to observe type ¢ñ and type ¢ó collagen, and immunohistochemistry was used to detect the expression of MMP-9. Results Compared with the normal group, the model group formed vulnerable plaques; compared with the model group, type ¢ñ collagen increased, type ¢ó collagen decreased (P£¼0.01), and the type ¢ó collagen/type ¢ñ collagen ratio decreased in the high- and low-dose traditional Chinese medicine groups ( P£¼0.01), MMP-9 expression decreased (P£¼0.01). Conclusion The Liuyu treatment method can stabilize vulnerable plaques, and regulating the content of type ¢ñ and ¢ó collagen and inhibiting the expression of MMP-9 may be one of its mechanisms.
