Abstract
Objective: To study the effect of curcumin against excessive deposition of extracellular matrix in bleomycin-induced pulmonary fibrosis in rats. Methods: 144 SD male rats were divided into sham operation group, model group, high-, medium- and low-dose curcumin groups, and prednisone acetate group, with 24 rats in each group. Rats in each group except the sham operation group were given a one-time infusion of bleomycin hydrochloride into the trachea, and the rats in the sham operation group were given a one-time infusion of an equal volume of normal saline into the trachea. Administration was started on the 1st day after modeling and continued until the day before the animals were sacrificed. The curcumin high, medium and low dose groups were 200, 100, 50mg.kg-1.d-1 respectively, the prednisone acetate group was 0.56mg.kg-1.d-1, the sham operation group and the model group were administered by gavage respectively. volume of saline. Eight animals in each group were randomly killed on days 7, 14, and 28, and the contents of hydroxyproline (HYP) in rat lung tissue and serum type III collagen, type IV collagen, laminin, and hyaluronic acid were respectively observed. Lung tissue was taken for HE and Mallory staining. Results: Curcumin can reduce the serum levels of collagen III, IV, laminin and hyaluronic acid in rats with pulmonary fibrosis, and reduce fibrous hyperplasia of lung tissue. Conclusion: It may exert its therapeutic effect on pulmonary fibrosis by reducing the extracellular matrix content in rats with pulmonary fibrosis.
